Rei Ayanami is one of NERV's primary pilots, and the one used in testing and calibrating the Evangelion units. Indeed, as the First Child, she's one of the only people who can pilot an Evangelion, with another Child in Japan only being discovered relatively recently. Presumably because she's often pushed to her limits and beyond as the only available pilot to NERV Japan, she's grown up quiet, withdrawn, and obedient, with little presence or sense of self-worth. She lives in a singularly barren apartment in Tokyo-3, and the only person to whom she ever seems to smile at is Commander Gendo Ikari.
